Eukaryotic histones are basic and water soluble nuclear proteins that form hetero-octameric nucleosome particles by wrapping 146 base pairs of DNA in a left-handed super-helical turn sequentially to form chromosomal fiber. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form the octamer; formed of two H2A-H2B dimers and two H3-H4 dimers, forming two nearly symmetrical halves by tertiary structure. Over 80% of nucleosomes contain the linker Histone H1, derived from an intronless gene, that interacts with linker DNA between nucleosomes and mediates compaction into higher order chromatin. Histones are subject to posttranslational modification by enzymes primarily on their N-terminal tails, but also in their globular domains. Such modifications include methylation, citrullination, acetylation, phosphorylation, sumoylation, ubiquitination and ADP-ribosylation.

Lane 1: 293T (Human embryonic kidney cells) cells transfected with PADI4
Lane 2: 293T cells transfected with PADI4 treated with 10 mM calcium chloride and 10 μM lonomycin for 2 hours

Lysates/proteins at 20 µg/Lane.
Exposure time: 4 seconds; ECL: K1801

Blocking: 5% NFDM/TBST, 1 hour at room temperature
Primary antibody: HA751893, 1/2,000 in primary antibody dilution buffer (K1803), overnight at 4 °C
Secondary antibody: Goat anti-Rabbit IgG-HRP (HA1001), 1/50,000 in 5% NFDM/TBST, 1 hour at room temperature

Immunoprecipitation (IP)

Histone H3 (citrulline R2) was immunoprecipitated in 0.2 mg 293T (Human embryonic kidney cells) cells transfected with PADI4 treated with 10 mM calcium chloride and 10 μM lonomycin for 2 hours cell lysate with HA751893 at 2 µg/10 µl beads. Western blot was performed from the immunoprecipitate using HA751893 at 1/1,000 dilution. Anti-Rabbit IgG for IP Nano-secondary antibody (NBI01H) at 1/5,000 dilution was used for 1 hour at room temperature.

Lane 1: 293T cells transfected with PADI4 treated with 10 mM calcium chloride and 10 μM lonomycin for 2 hours cell lysate (input)
